Functions and Effects:

L17E is a attenuated, cationic amphiphilic lysis (ACAL) peptide that can be used for the delivery of various macromolecules, including proteins, antibodies, and DNA nanostructures. L17E inserts into and disrupts membrane structures via electrostatic interactions, thereby facilitating endosomal escape. There is a strong correlation between the delivery efficiency mediated by L17E and the expression level of KCNN4—a gene encoding the calcium-activated potassium channel KCa3.1. Moreover, L17E can enhance cellular uptake of macromolecules by inducing micropinocytosis. L17E can be further optimized and improved through dimerization strategies or by combination with other delivery systems, such as nuclear localization signal peptides and cell-membrane-coated nanoparticles.
